       mRNA疫苗是一類新型核酸疫苗.相比于DNA疫苗,mRNA疫苗無需進入細胞核,無整合到基因組的風險,相對更安全.但mRNA自身穩定性差,易被體內外的核酸酶降解是制約其發展的瓶頸.因此,mRNA疫苗需要有合適的遞送載體將其遞送至體內,才能有更好的免疫效果.最早使用的脂質體遞送材料是陽離子脂質體。帶正電荷的陽離子脂質體與帶負電荷的 mRNA 通過經靜電作用聚集形成多層囊狀復合物,稱為脂質體復合物,復合物中包封的 mRNA 不易被 RNase 降解,能被成功遞送至細胞翻譯成功能性蛋白。陽離子聚合物具有濃縮 mRNA 的作用,mRNA 與聚合物濃縮后被陽離子脂質體包裹在囊狀空腔中則形成脂質體聚合物(lipopolyplex,LPR)。
